It has been three months since three days of arguments before the Supreme Court wrapped up on the Obama health law, and now the Justices will finally deliver their verdict.

But here is something important to remember - there is not just one health care case before the Supreme Court - there are three cases, and there are really four or five major decisions that must be made by the Justices.

What is not clear right is will there be one main decision covering those major points and all three cases? Or will there be different decisions with different majorities on the Obama health law?

* Case 11-393 is National Federal of Independent Business v. Sebelius - this case raises the issue of whether the entire law must be thrown out if the individual mandate is found to be unconstitutional.

* Case 11-398 is U.S. Department of Health and Human Services v. Florida - this is the basic case on whether Congress exceeded its powers by enacting the individual mandate which would force all Americans to purchase health insurance, and whether this case can even be brought at this point in time before the tax penalty for not buying insurance has been levied.

* Case 11-400 is Florida v. Department of Health and Human Services - this is Florida's challenge to provisions of the Obama health law that forces states to make health care changes in order to keep federal dollars for the Medicaid program.

So, maybe you can put together a flow of decisions like this:

1) Can the individual mandate be challenged before 2014?
2) If the answer is yes, is the individual mandate constitutional?
3) If the individual mandate is not constitutional, should the whole law be invalidated?
4) If the answer is that parts of the health law can survive, which ones?
5) And if some portions of the Obama health law survive, are the Medicaid provisions constitutional?

When you look at all of those possibilities, it makes sense that there is no clear answer on how this ruling will be presented.

Do you get one larger opinion in which a changing majority of Justices take different sides?

Soon after 10 am EDT on Tuesday morning, we will start to find out the answers on the Obama health law.

Programmable DNA scissors found for bacterial immune system

ScienceDaily (June 28, 2012) ? Genetic engineers and genomics researchers should welcome the news from the Lawrence Berkeley National Laboratory (Berkeley Lab) where an international team of scientists has discovered a new and possibly more effective means of editing genomes. This discovery holds potentially big implications for advanced biofuels and therapeutic drugs, as genetically modified microorganisms, such as bacteria and fungi, are expected to play a key role in the green chemistry production of these and other valuable chemical products.

Jennifer Doudna, a biochemist with Berkeley Lab's Physical Biosciences Division and professor at the University of California (UC) Berkeley, helped lead the team that identified a double-RNA structure responsible for directing a bacterial protein to cleave foreign DNA at specific nucleotide sequences. Furthermore, the research team found that it is possible to program the protein with a single RNA to enable cleavage of essentially any DNA sequence.

"We've discovered the mechanism behind the RNA-guided cleavage of double-stranded DNA that is central to the bacterial acquired immunity system," says Doudna, who holds appointments with UC Berkeley's Department of Molecular and Cell Biology and Department of Chemistry, and is an investigator with the Howard Hughes Medical Institute (HHMI). "Our results could provide genetic engineers with a new and promising alternative to artificial enzymes for gene targeting and genome editing in bacteria and other cell types."

Doudna is one of two corresponding authors of a paper in the journal Science describing this work titled "A programmable dual RNA-guided DNA endonuclease in adaptive bacterial immunity." The second corresponding author is Emmanuelle Charpentier of the Laboratory for Molecular Infection Medicine at Sweden's Ume? University. Other co-authors of the paper were Martin Jinek, Krzysztof Chylinski, Ines Fonfara and Michael Hauer.

Bacterial and archaeon microbes face a never-ending onslaught from viruses and invading circles of nucleic acid known as plasmids. To survive, the microbes deploy an adaptive-type nucleic acid-based immune system that revolves around a genetic element known as CRISPR, which stands for Clustered Regularly Interspaced Short Palindromic Repeats. Through the combination of CRISPRs and associated endonucleases, called CRISPR-associated -- "Cas" -- proteins, bacteria and archaeons are able to utilize small customized crRNA molecules (for CRISPR-derived RNA) to target and destroy the DNA of invading viruses and plasmids.

There are three distinct types of CRISPR/Cas immunity systems. Doudna and her colleagues studied the Type II system which relies exclusively upon one family of endonucleases for the targeting and cleaving of foreign DNA, the Cas9 proteins.

"For the Type II CRISPR/Cas system, we found that crRNA connects via base-pairs with a trans-activating RNA (tracrRNA), to form a two-RNA structure," Doudna says. "These dual RNA molecules (tracrRNA:crRNA) direct Cas9 proteins to introduce double-stranded DNA breaks at specific sites targeted by the crRNA-guide sequence."

Doudna and her colleagues demonstrated that the dual tracrRNA:crRNA molecules can be engineered as a single RNA chimera for site-specific DNA cleavage, opening the door to RNA-programmable genome editing.

"Cas9 binds to the tracrRNA:crRNA complex which in turn directs it to a specific DNA sequence through base-pairing between the crRNA and the target DNA," Doudna says. "Microbes use this elegant mechanism to cleave and destroy viruses and plasmids, but for genome editing, the system could be used to introduce targeted DNA changes into the genome.

Doudna notes that the "beauty of CRISPR loci" is that they can be moved around on plasmids.

"It is well-established that CRISPR systems can be transplanted into heterologous bacterial strains," she says. "Also, there is evidence to suggest that CRISPR loci are horizontally transferred in nature."

Doudna and her colleagues are now in the process of gathering more details on how the RNA-guided cleavage reaction works and testing whether the system will work in eukaryotic organisms including fungi, worms, plants and human cells.

"Although we've not yet demonstrated genome editing, given the mechanism we describe it is now a very real possibility," Doudna says.

This work was funded primarily by the Howard Hughes Medical Institute, the Austrian Science Fund and the Swedish Research Council.

Review: Superior Seafood | Restaurant Review | Gambit New ...

Superior Seafood & Oyster Bar is a sprawling place, and it needs to be. An upscale offshoot of the Superior Grill chain of Mexican cantinas, the restaurant holds down one corner of an important Uptown intersection, where streetcars rumble past and Mardi Gras parades make their sweeping turns. It's major real estate that calls for something significant.

??Superior Seafood also must be big to handle the many niches it's trying to cover, an undertaking that yields an unusual combination of an old French-Creole and modern family eatery, as if Galatoire's and Ralph & Kacoo's were combined. The result is a huge, pricey, busy, good-looking restaurant full of dichotomies, some of them fun (or at least useful), some of them just plain vexing.

??Big slabs of butter arrive with bags of hot French bread, though on two of four visits the bread was stale. The restaurant stocks a decent wine list, ranging from perfectly quaffable carafe wines to better selections. But behind the long zinc bar, daiquiri machines dispense frozen drinks. It's a come-as-you-are place, where reservations are accepted but rarely seem to be made and where service continues until midnight on weekends. A couple ordering the marquee dishes can easily spend $120 for dinner (prices come down a good deal at lunch).

??Quality varies widely, and it's hard to guess which dishes will satisfy. Price and complexity are not reliable indicators here. The $17 linguini with shrimp and pesto was a relative bargain on this menu and one of the better dishes I tried, as was a $27 entree of scallops, each crowned with a crisp edge and slivered garlic. Shrimp brochettes, filled with sausage and liquefied Monterey Jack and wrapped in charred bacon, were as delicious as they were indelicate.

??But the $29 redfish arrived so heavily salted it could've lasted until winter. Many dishes share the same sides, namely uniformly scooped portions of spicy maque choux and standard-issue whipped potatoes. These all but buried a blackened drum Napoleon, which collapsed under a goopy crawfish etouffee. A good crabmeat maison should not feature shredded lump crabmeat. Superior's is an orb that mixes shrimp and crab and heavily favors the shrimp mixed with strands of crab.

??Guests are paying for fine dining and getting something at least a full notch lower, but there are other considerations here. Families can bring wailing young children, and boy do they. Your cabal of visiting relatives all will be accommodated as readily here as at Superior Grill, a noted expert in crowd service, and everyone will find something to order somewhere on the menu.

??Superior Seafood gets some credit just for rescuing this address, a former Copeland's that sat blighted for years after Hurricane Katrina. If nothing else, that's worth a happy hour stop for some slurping and sipping at Superior Seafood's beautiful marble altar of an oyster bar or lunch out on the sunny patio as the streetcars roll past.

Pixar?s first heroine has been greatly anticipated for a long time, and she has finally arrived in ?Brave??a heartwarming, fable-like tale about courage and pride.

Sassy and as bold as her bright red hair, Scottish princess Merida (voiced by Kelly Macdonald) loves adventure, archery, and freedom. When she is told by her parents, Queen Elinor (Emma Thompson) and King Fergus (Billy Connolly), that she will soon be married off to a son of one of the kingdom?s lords, Merida storms out of the castle and attempts to search for her own fate.

Marriage is the last thing on Merida?s mind, even though her mother has been training her to be queen her entire life.

Things start getting interesting when she crosses paths with a suspicious old woodcarver who claims she can help Merida get her wish that her mother ?will change.?

?Brave? directors Mark Andrews and Brenda Chapman creatively insert plot twists that are sure to entertain and surprise the audience. This storyline is tightly wrapped around the mother-daughter relationship of Merida and the queen, sharing a few reminiscent qualities with ?Tangled.?

The stunning animations done by Pixar studios beautifully portray Scotland?s terrain and bring its people and history to life.

Merida is an immensely fierce and strong character. Her character is complex enough to be appreciated by an older audience but stubborn enough to appeal to younger audience members, who may likely relate to her ?That?s so unfair!? arguments with her mother.

Macdonald provides the perfect accent and energy as Merida?s voice. The entire vocal ensemble is very fitting, including Thompson as the mother struggling to break through the communication barrier between her and her daughter, and Connolly playing the easy-going parent.

Merida?s three younger, mischievous brothers don?t share much of a vocal presence, but their little gags and tricks provide great laughs from beginning to end.

The Scottish authenticity of the animations is breathtaking, and even the cultural jokes are done tastefully. The strong sense of community among the characters and their connection to their Scottish roots is very prominent.

A host of excellent qualities can be found in ?Brave??the action, the humor, the characters, the story, and the morality. This fantastic animated picture moves at a quick and steady pace, and it nails the humor without sacrificing the emotion and lessons about pride and family.

?Brave? can easily outshine any other animation feature for originality and heart, but may seem slightly underwhelming only when measured by the audience?s expectations of Pixar and alongside previous classics such as ?Toy Story? and ?Ratatouille.?

The Vampire Diaries hunk took to his Twitter to refute reports that he is "desperate" to portray Christian Grey, and to quell critics of his potential casting.

Somerhalder, 33, has been frequently talked about as a possible Christian not only due to his physical appearance, but his sexy, approachable demeanor.

Last week, he seemed to be full-on pushing for the Fifty Shades of Grey part, Tweeting at writer Bret Easton Ellis that he'd love Ellis to write the script for him.
